For over thirty years, the theophylline riboswitch has served as a cornerstone of synthetic biology, providing scientists with a programmable mechanism to turn genes ON and OFF using a chemical inducer. However, despite its widespread applications, the system has been limited by relatively moderate performance of its ligand inducers.
